mir-506 inhibits cell proliferation and invasion by targeting tet family in colorectal cancer

چکیده
objective(s): ten-eleven translocation (tet) family members have been shown to be involved in the development of many tumors. however, the biological role of the tet family and its mechanism of action in colorectal carcinogenesis and progression remain poorly understood. materials and methods:we measured the expression levels of tet family members in colorectal cancer (crc) specimens, in the corresponding normal tissues and in cell lines using quantitative real-time pcr (qrt-pcr) and in situ hybridization (ish). both the protein function and the protein-independent role of tets were investigated by cell viability assays and cell invasion assays using in vitro and in vivo models. results: we found that all three tet genes were strongly up-regulated at the transcript level in crc samples compared to matched normal tissues. the same results were observed in colorectal cancer cell lines. knockdown of tets by shtet1/2/3 showed that tet family members inhibited crc growth and metastasis. we showed that tet family member degradation by mir-506 inhibits cell proliferation and invasion in colorectal cancer. conclusion: through this study, we advance our understanding of the expression levels tets and mir-506 in crc and further clarify the internal regulatory mechanism of mir-506 by targeting tet during crc processes. these findings may contribute to a novel avenue for researching and developing targeted therapies for crc.

متن کاملMiR-141 inhibits colorectal cancer cell proliferation and invasion by targeting PRKAR1A
Dysregulation of microRNAs (miRNAs) has been associated with the initiation and progression of oncogenesis in humans. Growing evidence has indicated that abnormal expression of microRNA-141 was associated with tumorigenesis and carcinoma progression of various cancers. In the present study, the miR-141 expression was assessed in CRC cell lines and the normal colonic cell using RTq-PCR, which de...
متن کاملMiR-493 suppresses the proliferation and invasion of gastric cancer cells by targeting RhoC
Objective(s):MiRNAs have been proposed to be key regulators of tumorigenesis, progression and metastasis. However, their effect and prognostic value in gastric cancer is still poorly known. Materials and Methods: Gastric cancer cell lines were cultured. Tissue samples obtained from 36 gastric cancer patients were used for quantitative real-time PCR (qRT-PCR) analysis. The tissue microarrays (T...
متن کاملmiR-600 inhibits cell proliferation, migration and invasion by targeting p53 in mutant p53-expressing human colorectal cancer cell lines
Mutations of the tumor protein p53 gene, a tumor suppressor, are one of the most frequent genetic alterations observed in cancer. It has been reported that mutations in p53 result in the loss of wild-type p53 activity, and the gain of novel oncogenic properties that promote tumor growth and progression. Recent studies have demonstrated that a number of microRNAs (miRs) are involved in the post-...
متن کاملMiR-570 inhibits cell proliferation and glucose metabolism by targeting IRS1 and IRS2 in human chronic myelogenous leukemia
Objective(s): Chronic myelogenous leukemia (CML) is a chronic myeloproliferative disorder characterized by the accumulation of myeloid cells with a chromosomal translocation known as the Philadelphia chromosome. In this study, we investigated the roles of miR-570 in CML development. Materials and Methods: Expression of miR-570 in CML samples and cell lines was determined by qRT-PCR. Glucose upt...
